abstract

生物試料中の微細構造を蛍光標識し、可視化する蛍光バイオイメージング法は、近年の生物学研究において必要不可欠なツールとなっています。本法は、生物学研究者の要求に応える形で多様な技術発展を遂げてきました。中でも、レーザー光を試料上で走査し、各点における蛍光信号強度をPC上で再構築することで画像を取得するレーザー走査型蛍光顕微鏡は、1980 年代以降、飛躍的に普及しました。レーザー走査型蛍光顕微鏡の1つである共焦点顕微鏡は、光の波動性によって決定される回折限界に概ね相当した空間分解能を有することから、現在、様々な生命現象の詳細可視化に用いられています。近年の技術革新はさらに目覚ましく、様々な光学、物理学、化学の原理の応用により回折限界を打破した蛍光顕微鏡、いわゆる超解像顕微鏡が開発され、実用に至りつつあります。超解像顕微鏡法の確立に貢献した3名の研究者に2014年のノーベル化学賞が受賞されたことは、生物学研究者から大きな期待を象徴しているといえます。

本講演では、我々が新規に技術開発を行っている二光子励起蛍光顕微鏡について紹介します。二光子励起蛍光顕微鏡は、超短パルスレーザーを励起光源とし、発色団の二光子励起蛍光取得を前提としたレーザー走査型顕微鏡です。本法は、励起の局所性、高い深部到達性、低い侵襲性という生物学的可視化分析手法として、好ましい特徴を有しています。生命現象を可視化する際に、空間分解能、時間分解能、侵襲性、深部到達性と視野の広さは同等に重要です。我々は二光子励起蛍光顕微鏡について、超解像顕微鏡法の原理を応用することによる空間分解能の向上 [1] を、多点走査方式を適用することによる時間分解能の向上 [2] を、新規半導体レーザー光源の導入により深部到達性の向上を [3, 4] をそれぞれ達成しました。これらの顕微鏡システムの詳細について、可視化した作例とともに紹介したいと考えています。
